"THBD" - thrombomodulin in Homo sapiens
Nodes:
Network nodes represent proteins
splice isoforms or post-translational modifications are collapsed, i.e. each node represents all the proteins produced by a single, protein-coding gene locus.
Node Size
small protein node
small nodes:
protein of unknown 3D structure
large protein node
large nodes:
some 3D structure is known or predicted
Node Color
colored protein node
colored nodes:
query proteins and first shell of interactors
non-colored protein node
white nodes:
second shell of interactors
Edges:
Edges represent protein-protein associations
associations are meant to be specific and meaningful, i.e. proteins jointly contribute to a shared function; this does not necessarily mean they are physically binding each other.
Known Interactions
database edge
from curated databases
experiment edge
experimentally determined
Predicted Interactions
neighborhood edge
gene neighborhood
fusion edge
gene fusions
cooccurrence edge
gene co-occurrence
Others
textmining edge
textmining
coexpression edge
co-expression
homology edge
protein homology

THBDthrombomodulin; Thrombomodulin is a specific endothelial cell receptor that forms a 1-1 stoichiometric complex with thrombin. This complex is responsible for the conversion of protein C to the activated protein C (protein Ca). Once evolved, protein Ca scissions the activated cofactors of the coagulation mechanism, factor Va and factor VIIIa, and thereby reduces the amount of thrombin generated (575 aa)
Predicted Functional Partners:
F2
coagulation factor II (thrombin); Thrombin, which cleaves bonds after Arg and Lys, converts fibrinogen to fibrin and activates factors V, VII, VIII, XIII, and, in complex with thrombomodulin, protein C. Functions in blood homeostasis, inflammation and wound healing (622 aa)
        score_image score_image score_image   0.998
PROC
protein C (inactivator of coagulation factors Va and VIIIa); Protein C is a vitamin K-dependent serine protease that regulates blood coagulation by inactivating factors Va and VIIIa in the presence of calcium ions and phospholipids (461 aa)
        score_image score_image score_image   0.981
PF4
platelet factor 4; Released during platelet aggregation. Neutralizes the anticoagulant effect of heparin because it binds more strongly to heparin than to the chondroitin-4-sulfate chains of the carrier molecule. Chemotactic for neutrophils and monocytes. Inhibits endothelial cell proliferation, the short form is a more potent inhibitor than the longer form (101 aa)
        score_image score_image score_image   0.957
F5
coagulation factor V (proaccelerin, labile factor); Central regulator of hemostasis. It serves as a critical cofactor for the prothrombinase activity of factor Xa that results in the activation of prothrombin to thrombin (2224 aa)
          score_image score_image   0.926
PROS1
protein S (alpha); Anticoagulant plasma protein; it is a cofactor to activated protein C in the degradation of coagulation factors Va and VIIIa. It helps to prevent coagulation and stimulating fibrinolysis (676 aa)
          score_image score_image score_image 0.914
F10
coagulation factor X; Factor Xa is a vitamin K-dependent glycoprotein that converts prothrombin to thrombin in the presence of factor Va, calcium and phospholipid during blood clotting (488 aa)
          score_image score_image   0.905
JUN
jun proto-oncogene; Transcription factor that recognizes and binds to the enhancer heptamer motif 5’-TGA[CG]TCA-3’. Promotes activity of NR5A1 when phosphorylated by HIPK3 leading to increased steroidogenic gene expression upon cAMP signaling pathway stimulation (331 aa)
          score_image score_image   0.900
FOSL1
FOS-like antigen 1 (271 aa)
          score_image     0.900
PF4V1
platelet factor 4 variant 1; Inhibitor of angiogenesis. Inhibitor of endothelial cell chemotaxis (in vitro) (104 aa)
          score_image     0.900
CLEC4C
C-type lectin domain family 4, member C; Involved in antigen-capturing. Target ligand into antigen processing and peptide-loading compartments for presentation to T-cells. May mediate potent inhibition of induction of IFN-alpha/beta expression in plasmacytoid dendritic cells. May act as a signaling receptor that activates protein- tyrosine kinases and mobilizes intracellular calcium. Does not seem to bind mannose (213 aa)
