STRINGSTRING
STRING protein interaction network
Nodes:
Network nodes represent proteins
splice isoforms or post-translational modifications are collapsed, i.e. each node represents all the proteins produced by a single, protein-coding gene locus.
Node Color
colored nodes:
query proteins and first shell of interactors
white nodes:
second shell of interactors
Node Content
empty nodes:
proteins of unknown 3D structure
filled nodes:
a 3D structure is known or predicted
Edges:
Edges represent protein-protein associations
associations are meant to be specific and meaningful, i.e. proteins jointly contribute to a shared function; this does not necessarily mean they are physically binding to each other.
Known Interactions
from curated databases
experimentally determined
Predicted Interactions
gene neighborhood
gene fusions
gene co-occurrence
Others
textmining
co-expression
protein homology

nmt14-amino-5-hydroxymethyl-2-methylpyrimidine phosphate synthase; Responsible for the formation of the pyrimidine heterocycle in the thiamine biosynthesis pathway. Catalyzes the formation of hydroxymethylpyrimidine phosphate (HMP-P) from histidine and pyridoxal phosphate (PLP). The protein uses PLP and the active site histidine to form HMP-P, generating an inactive enzyme. The enzyme can only undergo a single turnover, which suggests it is a suicide enzyme. (346 aa)
Predicted Functional Partners:
thi2
Thiamine thiazole synthase; Involved in biosynthesis of the thiamine precursor thiazole. Catalyzes the conversion of NAD and glycine to adenosine diphosphate 5- (2-hydroxyethyl)-4-methylthiazole-2-carboxylic acid (ADT), an adenylated thiazole intermediate. The reaction includes an iron- dependent sulfide transfer from a conserved cysteine residue of the protein to a thiazole intermediate. The enzyme can only undergo a single turnover, which suggests it is a suicide enzyme. May have additional roles in adaptation to various stress conditions and in DNA damage tolerance.

thi4
Probable thiamine biosynthetic bifunctional enzyme; Condenses 4-methyl-5-(beta-hydroxyethyl)thiazole monophosphate (THZ-P) and 2-methyl-4-amino-5-hydroxymethyl pyrimidine pyrophosphate (HMP-PP) to form thiamine monophosphate (TMP). In the C-terminal section; belongs to the Thz kinase family.

 0.936
SPBP8B7.18c
Putative hydroxymethylpyrimidine/phosphomethylpyrimidine kinase 2; Catalyzes the phosphorylation of hydroxymethylpyrimidine phosphate (HMP-P) to HMP-PP, and of HMP to HMP-P. In the N-terminal section; belongs to the ThiD family.

 0.924
thi1
Thiamine repressible genes regulatory protein thi1; Transcription factor that activates the nmt1 promoter. Regulation of thiamine repressible genes. Positively regulates conjugation during meiosis.

 0.923
SPCC18B5.05c
Putative hydroxymethylpyrimidine/phosphomethylpyrimidine kinase C18B5.05c; Catalyzes the phosphorylation of hydroxymethylpyrimidine phosphate (HMP-P) to HMP-PP, and of HMP to HMP-P. Belongs to the ThiD family.

 0.922
SPBP8B7.17c
Putative hydroxymethylpyrimidine/phosphomethylpyrimidine kinase 1; Catalyzes the phosphorylation of hydroxymethylpyrimidine phosphate (HMP-P) to HMP-PP, and of HMP to HMP-P. In the N-terminal section; belongs to the ThiD family.

 0.917
cdc20
DNA polymerase epsilon catalytic subunit A; DNA polymerase II participates in chromosomal DNA replication; Belongs to the DNA polymerase type-B family.

      0.899
tnr3
Thiamine pyrophosphokinase; Involved in the regulation of thiamine metabolism, phosphate metabolism, mating and cell growth.

 0.889
pho4
Thiamine-repressible acid phosphatase pho4; May dephosphorylate thiamine phosphates; Belongs to the histidine acid phosphatase family.

 0.885
thi5
Thiamine repressible genes regulatory protein thi5; Transcription factor that activates the nmt1 promoter. Regulation of thiamine repressible genes. Negatively regulates conjugation during meiosis, by inducing negative regulators which delay conjugation. Involved in thi1 regulation.
